Glucose-dependent insulinotropic polypeptide is the other incretin alongside GLP-1, a 42-residue peptide from duodenal K cells that potentiates insulin release only when glucose is already elevated. DPP-4 cleaves it to the inactive GIP (3-42), so an assay that cannot separate intact from truncated peptide misreports incretin status — the central methodological problem in the field. Receptor-domain peptides and fragment analogues are used to study GIPR binding directly.
